Abstract

Structure-Based Genome-Editing Engineering In their Research Article (10.1002/sstr.202500674), Zhijia Yang, Hainan Zhao, and co-workers present Cas12Fold, a deep-learning framework tailored to Cas12 nucleases, that uses evolutionary signals and iterative structure-based alignment to resolve conformational complexity. This method outperforms general models in predicting key domains and states, accurately modeling enzymes such as Casλ. Models guide mutagenesis, optimizing Cas12j.4 efficiency and enabling CRISPR–Cas12 engineering.
