Abstract

Pirtobrutinib in frontline and relapsed settingsThe BRUIN-313 study of pirtobrutinib in treatmentnaïve older or unfit patients demonstrated early, durable responses, improving the survival in both IGHV unmutated and mutated patients compared to chemotherapy (HR 0.257, 95% CI 0.070, 0.934), despite a 52.9% crossover rate [2].Complementary results from BRUIN-314 demonstrated not only the non-inferiority of pirtobrutinib over ibrutinib but also its superiority in term of overall response rate and progression free survival (PFS), mainly in naive patients, as well as the better safety profile the led to a low rate of treatment discontinuation due to adverse events (7.9%, AEs) [3].
