Abstract

Chronic wounds remain a major clinical challenge and require advanced biomaterials that actively promote tissue regeneration. Here, elastin-like recombinamer (ELR)-based hydrogels functionalized with laminin-mimetic peptides (KAASIKVAVSADR and KPPFLMLLKGSTR) were developed using two complementary strategies: click chemistry grafting and recombinant genetic engineering. These bioengineered hydrogels provide a tailored microenvironment that enhances fibroblast and keratinocyte adhesion, proliferation, and migration, key processes in skin regeneration. Incorporation of the KPPFLMLLKGSTR peptide significantly improves keratinocyte adhesion and promotes re-epithelialization, whereas KAASIKVAVSADR supports cell adhesion, angiogenesis, and extracellular matrix remodeling. Additionally, the RGD sequence present in ELRs promotes fibroblast adhesion. The ELR matrix exhibits suitable mechanical properties, high water retention, and a porous structure facilitating nutrient exchange. In vitro assays confirmed excellent biocompatibility, high cell viability, robust adhesion, and enhanced cell migration compared with non-functionalized controls. Overall, this modular and scalable platform shows potential for next-generation wound dressings and future clinical translation.
