Come together: The BE-TILs—Efficient multiplex base editing of tumor-infiltrating lymphocytes
Abstract
The safe translation of multiplex gene-edited tumor-infiltrating lymphocytes (TILs) into an effective clinical product is like crossing a busy street: tumor excision, TIL isolation, precise gene editing, and rapid expansion form clearly defined, controlled steps while enabling forward progress, and come together in a unified, release-ready therapeutic (Figure 1). The study by Hafezi and colleagues,1 crossed the street using a scalable GMP-compatible TIL-manufacturing platform and adenine base editing (ABE) to achieve efficient dual knockout of the inhibitory receptors TIM-3 and TIGIT, while avoiding potentially hazardous DNA double-strand breaks (DSBs) associated with conventional CRISPR-Cas9 nucleases.
