LncRNA CXCL8-203 down-regulates IL-8 and enhance the radiosensitivity of Siha cells in cervical cancer
Abstract
To investigate the potential mechanisms by which IL-8 modulates radiosensitivity in SiHa cells. SiHa cells were selected for this study. Bioinformatic methods and RIP analysis were performed according to IL-8 upstream studies. qRT–PCR and Western blotting were used to detect gene and protein expression, respectively. The MTT assay and flow cytometry were conducted to assess cell viability and apoptosis, respectively, before and after irradiation. Co-immunoprecipitation (Co-IP) analyses of different proteins were performed before and after irradiation.IL-8 was regulated by long noncoding RNA (lnRNA) CXCL8-203. After lncRNA CXCL8-203 or IL-8 was silenced, the cell viability was decreased, and apoptosis was increased before and after irradiation. Apoptosis was reduced, and cell viability was increased by the application of Z-VAD-FMK after irradiation. According to the results of bioinformatic methods, qRT–PCR and Western blot analysis, Bax and Bcl-2 were selected for further evaluation. The interactions among Bax, Bcl-2 and IL-8 were lower after irradiation than before based on Co-IP. After Bax silencing, apoptosis was suppressed, and cell viability was increased; however, these effects were reversed by the IL-8 antibody. IL-8 promotes cell growth and inhibits apoptosis via Bax/Bcl-2 under the regulation of lncRNA CXCL8-203. IL-8 antibody application increases the radiosensitivity in SiHa cells.
