Abstract

Background: Depemokimab is the first ultra-long-acting biologic with enhanced IL-5 binding affinity, high potency and extended half-life, potentially enabling twice-yearly dosing. SWIFT-1/2 and ANCHOR-1/2 demonstrated depemokimab efficacy in patients with type 2 asthma characterised by blood eosinophils and patients with inadequately controlled chronic rhinosinusitis with nasal polyps (CRSwNP), respectively; in each, the proportion of patients having an adverse event (AE) was comparable across treatment arms. Aims: Assess depemokimab safety in pooled SWIFT/ANCHOR data. Methods: Patients aged≥12 (SWIFT)/≥18 (ANCHOR) years randomised 2:1/1:1, respectively, received depemokimab 100 mg subcutaneously or placebo, plus standard of care, every 26 weeks for 52 weeks. Results: Of 773 depemokimab and 517 placebo patients in the pooled SWIFT/ANCHOR safety population, 713 (92%) and 461 (89%) completed treatment/study, respectively. Mean exposure was 11.7 (SD 1.5) months in both treatment arms. Proportion of patients having on-treatment AEs/serious AEs (SAEs) was similar in both treatment arms with no fatal or treatment-related SAEs ([ Fig. 1 ]). Conclusions: Twice-yearly depemokimab is well-tolerated in patients with asthma or CRSwNP, supporting safety of the first ultra-long-acting biologic in these populations.
