Abstract

To the Editor: In the SOHO-01 study reported by Le et al. (Nov. 6 issue), 1 sevabertinib showed promising efficacy in patients with advanced HER2 -mutant non–small-cell lung cancer (NSCLC), particularly those with the YVMA subtype. Preclinical data 2 indicate that sevabertinib inhibits HER2 with mutant variants in intracellular or extracellular domains as well as wild-type HER2 yet has no activity in HER2-negative cells, which suggests a potential association with HER2 expression. An analysis of previous data regarding trastuzumab deruxtecan (formerly DS-8201) in HER2 -mutant NSCLC showed no correlation between response and HER2 expression status or expression intensity. 3 However, the SOHO-01 study lacks paired . . .
