Detection of seed-carried pathogen of soybean varieties Anjasmoro with the liquid assay method
Abstract
Soybean is one of the foods that can be processed into food that is consumed almost every day. Soybean seed production in Indonesia is still rare-because we often import soybeans from abroad. One of the reasons for the decrease in soybean seed production is the use of seeds infected with seed-borne pathogens. The use of seeds containing seed-borne pathogens will cause the quality of the seeds to decrease. To avoid losses due to decreased soybean seed production, one effort can be taken to identify seed-borne pathogens that cause disease in soybean seeds, so that the seeds produced will also be healthier and safer for cultivation. This study aims to: detect seed-borne pathogenic fungi in soybean seeds of the Anjasmoro variety using the liquid assay and identify the morphology of the seed-borne pathogenic fungi in soybean seeds of the Anjasmoro variety. This research will be conducted at the Seed Analysis Laboratory of Seed Technology Study Program, Lampung State Polytechnic from June to November 2023. Morphological identification experiments for seed-borne pathogenic fungi will be analyzed descriptively by observing the characteristics of each isolated fungus isolated from soybean seeds and compared with the literature. In the detection of pathogens using the liquid assay method, the growth medium for pathogenic fungi was isolated on PDA media. Variables observed included the morphology of pathogenic fungi visually using a magnifying glass and the mycelium shape of the fungi using a microscope. From the results of the research that has been carried out several conclusions were obtained, including the liquid assay method can be done to detect pathogens contained in soybean seed, two types of seed-borne pathogens were obtained, Sclerotium rolfsii and the bacterium Pseudomonas sp.
