Analysis of Gliricida genetic diversity using RAPD markers (random amplified polimorphic DNA)
Abstract
This research focuses on genetic variety and genetic characterization which is the initial stage of breeding program activities. The plant genetic material used in this molecular analysis activity was seven Gliricidia plant genotypes consisting of genotypes 38, 39, 40, 43, 46, 47, and 48 along with controls. The plant part used for molecular analysis activities is leaf tissue. Extraction of total plant genomic DNA was carried out using the small-scale CTAB method (miniprep) with minor modifications. Testing the quality of the isolated stock DNA solution was carried out using the electrophoresis technique. Amplification was carried out on a Thermocycler PCR machine. Data analysis was carried out using a scoring method for DNA bands that appeared in the results of 1.5% agarose gel electrophoresis. The scoring data was then analyzed using the UPGMA (Unweighted Pair-Group Method with Arithmetic)-SAHN (Sequential Agglomerative Hierarchial and Nested) program on NTSYS version 2.1 software. The Gliricidia accessions were clustered into two, the first cluster consisted of Control, 39, 40, 43, 46, and 47 which showed high genetic similarity values to Control. Meanwhile, the second cluster consists of genotypes 38 and 48. There is a possibility that genotypes 46 and 47 are the same genotype because they have a genetic similarity of 100%.
