Abstract

Analytical method development in liquid chromatography is fundamental to ensuring the quality, safety, and efficacy of pharmaceutical products. Recent advances in in-silico tools have introduced predictive and statistically driven strategies for systematic method optimization. Software platforms such as DryLab, Fusion, ACD/Labs, and Design Expert enable retention modeling, experimental design optimization, physicochemical property prediction, and statistical evaluation of critical method parameters. These tools facilitate the identification of method operable design regions (MODRs), improve robustness assessment, and reduce experimental workload. Integration of these computational approaches within the Analytical Quality by Design (AQbD) framework supports structured risk assessment, enhanced method understanding, and lifecycle management. In addition, analytical greenness and whiteness assessment metrics provide quantitative evaluation of environmental impact, analytical efficiency, and practical applicability. The combined implementation of in-silico modeling, AQbD principles, and sustainability assessment offers a comprehensive strategy for modern liquid chromatographic method development, emphasizing robustness, regulatory alignment, and environmental responsibility in pharmaceutical analysis.
