Abstract

Bicyclo[3.2.1]octanoid-type neolignans represent a class of natural compounds that have garnered growing interest due to their unique chemical structure and diverse bioactive properties. Found primarily in species of the Lauraceae, Magnoliaceae, and Piperaceae families, these neolignans possess a rigid, two-ring structure that provides molecular stability and potential for interaction with various biological targets. This text reviews various studies on the isolation, structural elucidation, and bioactivity of bicyclic [3.2.1]octanoid neolignans from different plant species. Several studies evaluated these compounds for biological activities, including inhibition of platelet-activating factor (PAF), anti-inflammatory, neurotrophic, and antimalarial properties. Thus, this review is essential for consolidating the knowledge on bicyclo[3.2.1]octanoid neolignans, highlighting their advances, challenges, and therapeutic potential. Additionally, it provides a solid foundation for guiding future research, encouraging investigations into new sources, synthetic optimizations, and clinical applications of these natural compounds.
