Abstract

Abstract The purpose of this study was to obtain putative mutants of Pothos tener Wall in vitro using gamma ray irradiation. This study consisted of two experiments, namely 1. the effect of BAP (benzyl amino purine), TDZ (thidiazuron) concentrations, and their interaction on Pothos tener Wall shoot multiplication in vitro, 2. the effect of gamma ray doses on Pothos tener Wall shoot growth in vitro. The method used was a completely randomized design (CRD) with two factors in experiment 1, namely BAP concentration (0 mg·L −1 , 3 mg·L −1 , and 5 mg·L −1 ) and TDZ concentration (0 mg·L −1 , 0.1 mg·L −1 , and 0.3 mg·L −1 ). Experiment 2, one-factor CRD, namely the dose of gamma ray radiation at (0 Gy, 5 Gy, 10 Gy, 15 Gy, 20 Gy). The results showed that BAP concentrations of 0 mg·L −1 and 3 mg·L −1 with the addition of TDZ concentration resulted in an increase in shoot height and an increase in the number of leaves that were not significantly different, but at a BAP concentration of 5 mg·L −1 with the addition of TDZ concentration, it could reduce the increase in shoot height and an increase in the number of leaves. The addition of radiation doses (gamma rays) starting from 10 Gy significantly reduced the percentage of explants that remained green.
