Quantitative holo-tomographic flow cytometry: unlocking label-free lysosomal biomarkers for LSDs
Abstract
Lysosomal Storage Diseases (LSDs) demand accurate lysosome visualization for understanding disease progression. We demonstrate that Holo-Tomographic Flow Cytometry (HTFC) enables 3D imaging of lysosomal compartments in single live cells. HTFC is an innovative, label-free tool for the visualization and quantification of subcellular compartments, enabling high-content and high-throughput analysis. This breakthrough allowed us to identify novel biomarkers of lysosomal accumulation and quantitatively correlate 3D lysosomal architecture with therapeutic efficacy. Our work significantly advances lysosomal research, paving the way for improved LSD diagnostics and targeted therapies.
