Figure S4 from RAS(ON) Multiselective Inhibition Drives Antitumor Immunity in Preclinical Models of NRAS-Mutant Melanoma
Abstract
Supplemental Figure 4: Complete responses to RMC-7977 are dependent upon CD4+ and CD8+ T cells. (A) Tumor growth and survival curve of syngeneic mouse model of OSUMMER.13 cell line. Control group was treated with vehicle (P.O., q.d.), Anti-CD4 group was treated with Anti-CD4 antibody (1µg/µL, I.P., every 4 days), Anti-CD8 group was treated with Anti-CD8 antibody (1µg/µL, I.P., every 4 days), Anti-CD4 + RMC-7977 was treated with Anti-CD4 antibody (1µg/µL, I.P., every 4 days) and RMC-7977 (25 mg/kg, P.O., q.d.); Anti-CD8 + RMC-7977 was treated with Anti-CD8 antibody (1µg/µL, I.P., every 4 days) and RMC-7977 (25 mg/kg, P.O., q.d.); and RMC-7977 group was treated with only RMC-7977 (25 mg/kg, P.O., q.d.) for 7 weeks. Control n=5M, Anti-CD4 n=5, Anti-CD8 n=5M, Anti-CD4 + RMC-7977 n=5M, Anti-CD8 + RMC-7977 n=5M and RMC-7977 n=5M. Plots correspond to individual mouse. (B) Total number of tumor infiltrating CD4 and CD8a T cells as measured by flow cytometry. Flow cytometry analysis confirmed the depletion of both CD4+ and CD8+ T cells in the depleted tumors.
