Next-Generation Immune Checkpoint Inhibitors in Upper Gastrointestinal Cancers: The Role of TIGIT
Abstract
T-cell immunoreceptor with Ig and ITIM domains (TIGIT) is a biologically rational target in upper gastrointestinal cancers, in which coexpression with PD-1 and abundant CD155 reinforces T-cell exhaustion and immune resistance. However, despite promising phase 2 signals, TIGIT blockade has not yet delivered consistent phase 3 benefit, and its additive value beyond PD-1 inhibition remains unknown in unselected populations.
