Abstract

The chapter delves into the transformative approach of quality by design (QbD) in drug formulation. Moving beyond traditional, reactive methods, QbD emphasizes a science-based, proactive strategy to ensure consistent, high-quality medications. It starts with clearly defined goals (desired therapeutic effect, safety profile) and identifies critical quality attributes (CQAs) of the final product. Risk assessment pinpoints potential areas of variability, allowing formulation design within a defined “design space” where CQAs remain optimal. Control strategies ensure process consistency throughout manufacturing. This chapter explores key tools such as design of experiments (DoE) for optimizing formulations, process analytical technology (PAT) for real-time monitoring, and multivariate data analysis for a deeper understanding. Practical applications across various dosage forms (oral, parenteral, topical) are showcased, along with successful case studies. Challenges and opportunities are discussed, including regulatory considerations, data management, and future advancements in QbD technologies. Ultimately, this chapter highlights how QbD empowers the pharmaceutical industry to deliver safe, effective, and consistently high-quality drugs, benefiting patients and healthcare professionals alike.
