Abstract

The cellular process of carcinogenesis greatly depends on Extrachromosomal DNA (ecDNA) since it offers cancer cells with three critical oncogenic qualities, including gene amplification increase, drug resistance development, and tumor cell heterogeneity maintenance. Studies employing sophisticated imaging tools provide better characterization of cancer cell ecDNA, allowing researchers to find new knowledge regarding its biological workings and clinical implications. This study includes a complete evaluation of recent methodologies utilized for ecDNA detection and analysis through fluorescence in situ hybridization (FISH), DNA sequencing protocols, and both super-resolution microscopy and electron microscopy high-resolution imaging tools. Precise localization and functional investigation of ecDNA in tumor microenvironments became achievable because of single-cell and spatial transcriptomics combinations. The study offers assessments of computational strategies for sequencing data analysis and machine learning applications dedicated to ecDNA identification. The chapter goes into the clinical significance of ecDNA because it describes its function in cancer progression and medication resistance potential while discussing prospective therapy applications based on ecDNA detection. A study of how ecDNA emerges and is defined in cancer cells will enable the development of focused disease-fighting therapies that precisely target ecDNA cancer propagation.
