Abstract

Crude extracts from natural products (NPs) continue to offer many bioactive compounds, which have essential uses in medicine, agriculture, and biotechnology applications. The conventional screening procedures cannot fully exploit the chemical diversity present in extracts due to the complexity of samples, together with limited quantitative techniques. This chapter covers innovative approaches that integrate synthetic biology, ultra-high-resolution analysis, artificial intelligence (AI) platforms, and bioengineering tools that manage systems to improve the discovery of NPs. Advanced green extraction technologies, including supercritical fluid and microwave-assisted extraction, opеrаtе for efficient and environmentally friendly compound ехtractіon using AI-operated solvent systems. Analytical platforms such as UHPLC–QTOF–MS, Orbitrap MS, and cryogenic NMR (cryo-NMR) offer unprecedented precision in real-time compound detection and structural elucidation. The bioactivity prediction, along with dereplication techniques, gained momentum because deep learning and generative algorithms operate within GNPS and NPClassifier platforms. Lab-on-a-chip, along with microfluidic devices, functions through AI commands to carry out rapid screenings at reduced material volume rates and real-time assay modulations. Bioassays of organ-on-chip technology and CRISPR-based biosensors allow researchers to conduct simultaneous high-content screening of phenotypic analysis at high resolution. Synthetic biology combines biosynthetic gene clusters (BGC) refactoring and metabolic engineering approaches to awaken inactive gene clusters, which results in a more diverse chemical composition of crude extract libraries. Self-driving laboratories linked with cloud-based analytics work toward developing autonomous discovery systems that aim to establish sustainable operational standards for future applications. This chapter envisions a new era in the discovery of NPs, built on ethics, innovation, and interdisciplinary cooperation, urging a collective effort from various scientific communities to explore the untapped potential of nature&s;s biochemical potential.
