Abstract

Recently, synthetic biologists have focused on RNA-based genetic circuits and regulatory mechanisms. This chapter compares RNA-based systems to DNA-based systems and their potential benefits. New RNA regulatory mechanisms like STARs, toehold translational switches, and CRISPR systems like CRISPRi allow scientists to control gene expression and design intricate genomic circuits. The chapter also discusses post-transcriptional gene regulatory circuits using mRNA switches and synthetic circuits’ complicated host–organism interactions. Gene therapy, microbiome engineering, agriculture, and industry can use genetic circuits for prediction. This chapter also discusses software that can examine and use RNA circuit components. RNA-based genetic circuits and regulatory mechanisms have grown rapidly and are important in synthetic biology. This chapter covers their growth and usage.
