Design of Riboswitches and Their Uses
Abstract
Riboswitches are structured non-coding RNA elements composed of an aptamer domain and an expression platform. The former acts as a ligand receptor, and the latter transduces the signal to regulate gene expression. Its inherent capability to function as a standalone regulatory element cryptically resembles a bridge between the ancient hypothetical RNA world and the modern central dogma of life. This chapter provides a comprehensive view of the ability of various design approaches inspired by natural riboswitches to elicit gene response. Riboswitch design strategies were explored toward mechanisms of transcription–translation regulations, biosensing, and coupling with ribozymes, each optimized through distinct approaches like SELEX, rational design, in silico modeling, and high-throughput screening. This chapter provides a structured overview of the various design approaches of riboswitches based on their mechanisms of action. In addition, it details the significant advances in synthetic riboswitch engineering, positioning riboswitches as versatile, adaptable tools for application in diverse fields such as metabolic engineering, synthetic biology, environmental monitoring, therapeutics, and diagnostics.
