Abstract

Synthetic biology is a multidisciplinary field which focuses on applications of bioengineering for new functions, rerouting of metabolic pathways, and reorganization of genomes for entire organisms. Synthetic biomolecular devices can sense and manipulate cellular physiology; they advance our understanding of natural biomolecular networks. RNA’s molecules offer flexible, customizable, consistent, reliable, and straightforward alternative to proteins for building genetic circuits. Riboregulators are RNA-based compounds which regulate gene expression. Eukaryotic gene regulation is significantly more complex than prokaryotic regulation which involves five levels: epigenetic, transcriptional, post-transcriptional, translational, and post-translational. Regulatory RNAs play an important role in regulating gene expression, which includes the transcription step to the translation and function of proteins in prokaryotes and eukaryotes. They are divided into three sub-families: riboswitches, 5′ UTRs, and short regulatory RNAs (sRNA), or antisense RNAs (asRNA). The main category is sRNAs, which are often encoded by distinct genes and typically function in trans, repressing target mRNAs post-transcriptionally by binding. Riboswitches are significant post-transcriptional regulators, found within the 5′ UTR of mRNA at the transcriptional and translational levels. sRNA, or antisense RNAs, regulate the efficiency of transcription, RNA stability, and the initiation of translation. Because of riboswitch affinity and selectivity, sRNAs are of the utmost interest for antibiotic development. These are interesting targets because they are associated with regulating metabolic pathways and stress responses, which are crucial for the development of biosensors, bacterial growth regulators, and therapeutic targets. Synthetic riboswitches can be constructed by assembling aptamers and regulatory domains. RNA thermometers play a vital role in the industrial control of bioprocesses, and hybrid regulatory devices have multifaceted potential in controlling the transcription and translation of target genes. Thus, RNA regulators provide a wide domain of applications, including metabolic engineering and therapeutic advancement.
