Abstract

In the next decade, metabolic diseases are projected to afflict one billion people, becoming the world’s top noncommunicable disease by 2030. Epigenetics plays a crucial role in regulating gene expression by modifying the chromatin structure and DNA methylation patterns. It has been intricately linked to the pathogenesis and susceptibility of a spectrum of metabolic disorders, including obesity, diabetes, nonalcoholic fatty liver disease, gout, osteoporosis, hypothyroidism, and hyperthyroidism. These associations underscore the intricate interplay between epigenetic modifications and the development of metabolic illnesses, revealing promising 278 avenues for further research and potential therapeutic interventions. Epigen-etic mechanisms, encompassing DNA methylation, histone modification, chromatin remodeling, and noncoding RNA processes, exert multifaceted influences on the development of metabolic diseases. These mechanisms are subject to genetic and nongenetic factors, including the aging process, dietary choices, and physical activity, collectively contributing to an individual’s phenotype formation. Recognizing the implications of epigenetics in the clinical context, such as through epigenetic biomarkers, epigenetic pharmaceutical interventions, and epigenetic editing, holds promise for diagnosing and treating metabolic disorders. This chapter provides an overview, commencing with the historical background of epigenetics and tracing critical milestones from the inception of the term “epigenetics.” It also outlines research methodologies employed in epigenetics and delineates the four principal epigenetic modulation mechanisms. Furthermore, the chapter synthesizes existing knowledge on epigenetic mechanisms relevant to metabolic diseases and elucidates the intricate interplay between epigenetics and genetic and nongenetic determinants.
