Abstract

This protocol details the preparation of 10 µm histological sections from paraplast-embeddedrat ovarian tissueusing a rotary microtome, followed by hematoxylin–eosin staining and permanent mounting. Key steps include sectioning, slide preparation, deparaffinization, staining, dehydration, clearing, and mounting. The resultingrat ovariansections exhibit strong tissue adherence, uniform staining, and clear nuclear-cytoplasmic contrast, making them suitable for routine histological examination and quantitative morphometric analysis.
