Evaluation of High-Throughput Platform for Wild Bird-Mediated Bloodborne Pathogens
Abstract
Wild birds migrating between countries can carry ticks or tick-borne pathogens, making them potential vectors for disease spread. This study evaluated and compared the performance of high-throughput platform (HTP)-based diagnostic techniques for four bloodborne pathogens potentially transmitted by wild birds. After evaluating the performance (limit of detection [LOD], coefficient of determination [R 2 ], PCR efficiency) of the selected singleplex qPCR assay for each pathogen, each assay that met the criteria was selected and examined to verify its performance on the HTP-based chip. The test results indicated that the singleplex qPCR assay had an LOD of 1 × 10 1 -1 × 10 2 molecules/µL, R 2 greater than 0.98, and acceptable PCR efficiency, comparable to the performance standards for conventional multiplex qPCR assay on HTP-based chips. The four qPCR assays investigated in this study are expected to be included as individual assays in HTP-based chips and will be utilized in a national-scale screening system for bloodborne infectious diseases in wild birds.
