BIOAVAILABILITAS DAN ADMET
Abstract
Bioavailability and ADMET evaluation are essential components of modern drug discovery, as they determine whether a compound can reach systemic circulation, interact effectively with its target, and remain safe for use. Bioavailability is influenced by physicochemical properties, formulation, and physiological factors, while drug-likeness rules such as Lipinski’s and Veber’s provide early screening criteria. ADMET analysis encompasses absorption, distribution, metabolism, excretion, and toxicity, offering a comprehensive view of a drug’s pharmacokinetic and safety profile. Early in silico prediction using computational tools allows inefficient or toxic compounds to be eliminated at the initial stages of development. Consequently, integrating bioavailability and ADMET assessment significantly improves the efficiency, cost-effectiveness, and success rate of drug development.
