Intestinal Microbiota Simulator for Clostridium difficile Recurrence Modelling
Abstract
A full-stack computational simulator modelling Clostridioides difficile infection (CDI) recurrence using an 11-species Lotka-Volterra ODE system with Monod kinetics and a multi-axis growth modifier layer (bile acid resistance, butyrate score, substrate availability, pH, oxygen). The model implements a Toxin/Damage axis (TcdA + TcdB) and a Monte Carlo pipeline (N=300, FNV-1a substream RNG) for probabilistic recurrence estimation across five therapeutic scenarios: S1: Vancomycin (rCDI reference: 27.3%) S2: Fidaxomicin (23.0%) S3: Bezlotoxumab (21.7%) S4: Faecal Microbiota Transplant — FMT (7.3%) S5: VOWST oral microbiome therapy (11.0%) The interaction matrix (α_ij, 59 entries) was calibrated through 8 formal sessions with 18 documented structural decisions. Sensitivity analyses confirm that the therapeutic gradient S1>S2>S3>S5>S4 is preserved under ±50% global α scaling and breaks at F=2×. The platform includes a React + TypeScript frontend, Express API server, PostgreSQL persistence, and an LLM Society layer for agent-based narrative exploration.
