Abstract

The extract of Datura metel leaves is a natural source of secondary metabolites with potential anticancer properties through the inhibition of cell proliferation and induction of apoptosis. This study aimed to evaluate the anticancer activity of Datura metel leaf extract against breast cancer cells using an in vitro approach. Ethanolic extract of the leaves was obtained by maceration and tested for cytotoxic activity using the MTT assay on MCF-7 breast cancer cells. The results demonstrated that the extract significantly reduced cell viability with an IC₅₀ value within a relevant concentration range as a potential herbal anticancer candidate. Morphological observations revealed structural changes such as cytoplasmic shrinkage and nuclear fragmentation, indicating apoptosis induction. Furthermore, the presence of alkaloids, flavonoids, and phenolic compounds in the extract is suggested to play a crucial role in cytotoxic mechanisms through oxidative stress induction and cell cycle arrest. These findings support the potential of Datura metel leaves as a natural phytochemical source for anticancer development; however, further studies are required to explore detailed molecular mechanisms and in vivo evaluation to ensure safety and efficacy. Therefore, Datura metel leaf extract holds promise as a complementary therapy in breast cancer treatment.
