Pegcetacoplan in Geographic Atrophy: Mechanistic Rationale, Clinical Evidence, Safety Profile, and Future Therapeutic Landscape - A Structured Narrative Review
Abstract
This project aims to conduct a structured narrative review of pegcetacoplan for the treatment of geographic atrophy secondary to age-related macular degeneration. The review will synthesize current knowledge regarding complement pathway involvement in disease pathophysiology, pharmacologic rationale for C3 inhibition, pivotal clinical trial data (including OAKS and DERBY), safety considerations, risk of exudative conversion, and emerging long-term extension findings. The review will also critically compare pegcetacoplan with other complement inhibitors and discuss future therapeutic implications.
