Abstract

This is the supplementary data for:Modular universal rbcL primers for (meta)barcoding: a systematic case study clarifying the identity of the marine macroalga Corallina armata (Rhodophyta: Corallinales)AL van der Reis1*, MA Sewell1, RM Wade2, and WA Nelson1,31 School of Biological Sciences, University of Auckland, New Zealand 2 Department of Biological Sciences, California State Polytechnic University, Arcata, California, USA3 Auckland War Memorial Museum Tāmaki Paenga Hira, Auckland, New Zealand *Corresponding author: aimee.vdreis@auckland.ac.nzThe material provided is split into two components - the development of the modular universal *rbc*L primers and the *Corallina armata* case study. There are additional README files within the 'Github.zip' folder to provide guidance for the various scripts. The subdirectories within the main 'Primer_dev' folder are labeled according to the appropriate scripts. The subdirectories within the main 'Corallina_armata.zip' folder are labeled according to their purpose and typically link back to the R code.* *'Sequence_processing.zip' is the output needed from the QIIME and BLAST results. *'*Geneious_files' have the various alignments and the subsequent trimming the was done for the various primer pairs. Files and variables:SUPP_Corallina_armata.xlsx - Article supplementary tables and figuresPrimer_dev.zip - Data (incl. Geneious files) and R projectCorallina_armata.zip - Data (incl. Geneious files) and R projectGithub.zip - ScriptsThe Geneious files need to be viewed using Geneious ([https://www.geneious.com/](https://www.geneious.com/)). The free/trial version will allow viewing, but will be limited in other functions. The R projects can be opened using R studio ([https://posit.co/download/rstudio-desktop/](https://posit.co/download/rstudio-desktop/)).Other files can be opened with most standard text editors, PDFs on web browsers, and jpegs in image viewers.
