Abstract

Background: Diarrhoea is a common gastrointestinal disorder and the leading cause of morbidity and mortality, particularly among children under five. Conventional treatments have (large) limitations with adverse effects. Ayurveda offers various alternative drugs that are effective for preventing diarrheal episodes. Rasa Parpati is traditionally employed for gastrointestinal disorders, but its antidiarrheal efficacy, particularly with Bilwa (Aegle marmelos), remains unverified. Aim: This study aims to analyse Tripatra Parpati and evaluate its acute toxicity and antidiarrheal activity in Wistar albino rats. Methods: The antidiarrheal potential was assessed using a castor oil-induced diarrhoea model, comparing Tripatra Parpati with Rasa Parpati and Bilwa. Parameters evaluated included diarrhoea onset and defecation percentage inhibition. An acute oral toxicity study was conducted following OECD guidelines 425. HPTLC, FTIR, XRD and SEM-EDAX were employed to assess the physico-chemical structures and components. Results: Tripatra Parpati was found safe at 2000 mg/kg and significantly reduced diarrhoea severity and frequency compared to control groups, with statistically significant differences. Conclusion: These findings support Tripatra Parpati as a safe and effective antidiarrheal formulation, particularly beneficial in resource-limited settings. Major Findings: Tripatra Parpati displayed stable physicochemical features and remained safe even at higher doses of 2000mg/kg during toxicity testing. The formulation showed notable antidiarrheal action, achieving around 61% inhibition in castor oil–induced diarrhoea, indicating its therapeutic potential with no significant adverse effects.
