Abstract

This study aimed to evaluate the ameliorative effect of platelet-rich plasma (PRP) against nephrotoxicity induced by paclitaxel (PTX). Rats were randomly divided into 4 groups: control group, PRP group, PTX group and PRP+PTX group. Rats of the PRP group received Intra Peritoneal (IP) injection of PRP (0.5 ml/kg. b.wt.) twice weekly. Rats of the PTX group received IP injection of PTX (7.5 mg/kg. b.wt.) once a week on days 0, 7, 14, 21 and 28. PRP+PTX group received IP injection of PTX (7.5 mg/kg. b.wt.) on days 0, 7, 14, 21 and 28, and IP injection of PRP (0.5 ml/kg. b.wt.) twice weekly. Administration of PTX significantly increased the serum levels of urea and creatinine. There was a significant increase in the serum level of malondialdehyde (MDA) and a significant decrease in the serum level of glutathione peroxidase (GPx) in PTX group. Histopathologically, PTX induced severe histological alterations in kidneys, including congested capillaries, glomerulonephrosis, severe vacuolation of renal tubules, necrosis of renal tubular epithelium, cystic formation, hyaline casts and perivascular mononuclear cellular infiltration. Administration of PRP restored the normal level of serum creatinine, numerically decreased the serum level of urea and induced a significant improvement in renal tissue. In conclusion, administration of PTX induced severe nephrotoxic effects, which can be manifested biochemically and histopathologically. Co-administration of PRP ameliorated the renal histopathological changes induced by PTX, which may be related to growth factors released by PRP.
