Emerging Strategies in Nanomedicine for Cancer Therapy: Overcoming Obstacles and Examining Possibilities
Abstract
Introduction: Cancer remains one of the world's leading causes of death, and traditional treatments such as radiation and chemotherapy often have limitations, including systemic toxicity, low selectivity, and multidrug resistance. Nanomedicine has emerged as a transformative strategy by utilizing nanoscale materials to enhance drug delivery, improve tumor targeting, and reduce side effects. Recent advances in nanocarriers, including metalbased nanostructures, dendrimers, liposomes, and polymeric nanoparticles, have shown considerable promise in overcoming pharmacokinetic and biological barriers. Methods: This study focuses on novel nanotechnological approaches in cancer treatment and provides a comprehensive analysis of recent peer-reviewed publications from 2018 to 2025. Information was gathered from databases such as Web of Science, PubMed, and Scopus, with an emphasis on research addressing stimuli-responsive release mechanisms, targeted delivery, and regulation of the tumor microenvironment to overcome pharmacokinetic barriers. Results: The results indicate that multifunctional nanoplatforms, incorporating components for imaging, therapy, and targeting, achieve greater treatment precision. Surface modification with ligands and antibodies enhances selective accumulation in tumor tissues through receptor-mediated endocytosis. Furthermore, nanocarriers responsive to pH, temperature, and redox conditions demonstrate improved drug release profiles and reduced systemic toxicity. Discussion: Despite encouraging results, clinical translation faces challenges related to large-scale synthesis, stability, biodistribution, and regulatory approval. Emerging developments, such as immune-modulating nanoparticles, hybrid nanocomposites, and personalised nanomedicine, are helping to bridge the gap between successful experimental outcomes and practical applications. Conclusion: An important advancement in oncology is nanomedicine, which offers innovative approaches for safe, effective, and targeted cancer treatment. To fully realise its therapeutic potential, further multidisciplinary research and clinical validation are essential.
