Abstract

Imidazo[1,2-a]pyridine, a privileged fused heterocyclic framework has attracted significant attention in medicinal chemistry due to broad pharmacological relevance and its structural diversity. The review compiles recent advancements in the medicinal potential by the derivatives of imidazo[1,2-a]pyridine. Numerous studies have presented the potential of this scaffold as anticancer, antimicrobial, anti-inflammatory, central nervous system (CNS), and antiprotozoal agents, with several compounds showing potency comparable to standard drugs. Imidazo[1,2-a]pyridine derivatives have been reported to act through diverse mechanisms, including enzyme inhibition (CDKs, PI3K?, HDACs, cholinesterases), modulation of signaling pathways (Wnt/?-catenin, NF-?B, STAT3), and receptor targeting (GABA-A, adenosine A1). Overall, the therapeutic versatility of the imidazo[1,2-a]pyridine framework has been emphasized in the review which also underscores its promise as a valuable lead framework for future drug discovery and development.
