Abstract

The histone demethylase KDM6B, expressed in cranial neural crest-derived dental mesenchyme, acts as a key epigenetic regulator coordinating sensory innervation during tooth root development. Loss of KDM6B impairs trigeminal axon entry and branching into the dental pulp by reducing the expression of the BMP antagonist Bambi, leading to overactivation of BMP signaling and suppressed NGF expression, which hinders sensory axon extension.
