Rational Design of a μ-Opioid Receptor Agonist with Integrated Calcium Channel Modulation: Computational Insights
Abstract
This purely computational study reports the in silico design and preliminary evaluation of a novel μ-opioid receptor agonist scaffold with potential dual modulation of calcium channels.[1,2,3]. Docking studies were performed on the deacetylated (active) form of the ligand, revealing favorable binding to μ-opioid receptor residues critical for activation, including hydrophobic contacts with TRP242, ILE245, VAL249, and key hydrogen bonds and a stabilizing salt bridge with HIS246 [1,2]. Interaction profiling against a voltage-gated calcium channel (8IF3) showed multiple hydrophobic and hydrogen-bond contacts, suggesting potential dual-target engagement [3,4]. Physicochemical and ADMET analysis identified limited lipophilicity, raising concerns for blood–brain barrier penetration [5]. To address this, a prodrug approach via selective acetylation of hydroxyl groups is proposed to enhance CNS penetration; enzymatic hydrolysis in situ is expected to release the active, deacetylated ligand capable of receptor engagement [5,6]. These results establish a foundation for further computational and experimental studies toward multifunctional analgesic agents [1,2,3].
