Formulation and evaluation of chronotherapeutic based tablet in tablet of paracetamol and baclofen
Abstract
The chronotherapeutic drug delivery systems are such that drugs are released in synchrony with the circadian rhythms of the body to achieve maximum therapeutic effect and minimum side effects. Various disorders such as pain, muscle spasticity exhibit time - dependent variation of the symptoms, which necessitates the use of control and programmed drugs release. The current study will endeavor to prepare a chronotherapeutic tableting - in - tablet formulation comprising of baclofen as the central tablet and paracet amol as the peripheral tablet. To obtain an initial immediate release of paracetamol to treat pain rapidly followed by a lag time and later controlled release of baclofen, the tablet - in - tablet method was used. The excipients were chosen appropriately in or der to achieve good flow characteristics, compressibility, and mechanical strength of the tablets. To ascertain the expected pattern of drug release, the formulated pills were assessed according to the physicochemical parameters and the in vitro drug relea se activity. The findings showed that the designed system was able to offer time - controlled and sequential drug delivery in line with the chronobiological needs. These tablets in therapeutic results.
