Abstract

This thesis explored genomic changes in human mesenchymal stem cells (hMSCs) following extended in vitro expansion and modification of the local microenvironment via the heparan sulfate proteoglycans. Using advanced next generation sequencing and epigenetic profiling techniques, the project determined transient gene expression and methylation changes in hMSCs as potential targets to enhance their therapeutic capacity. As MSCs are key candidates for stem cell-based therapies, these findings provide progress in advancing regenerative medicine applications for a range of conditions including neurodegenerative disorders.
