Immunoinformatics-Driven Design of a Multi-Epitope mRNA Vaccine Against Plasmodium vivax Reticulocyte-Binding Protein 3
Abstract
This work presents the computational design of a multi-epitope mRNA vaccine targeting Plasmodium vivax reticulocyte-binding protein 3 (PvRBP3), a key antigen involved in red-blood-cell invasion. Using immunoinformatics tools, I identified 12 MHC-I and 8 MHC-II T-cell epitopes with strong predicted HLA binding and broad population coverage (≈95%). These epitopes were assembled into a codon-optimized mRNA construct incorporating proven design elements including signal peptide, optimized UTRs, N1-methylpseudouridine modification, and LNP delivery. The report also outlines manufacturing steps, preclinical evaluation strategy, and clinical pathway considerations. This design provides a theoretical vaccine candidate and a reproducible pipeline for future malaria vaccine development
