Constraint-Induced Coherence for Early Detection of Protein Misfolding Cascades: A Monograph
Abstract
A 112-page monograph establishing that protein misfolding detection can be made mathematically deterministic. We prove that on a simply connected profinite site (π₁ = 0), all conformational trajectories converge to a unique fixed point, eliminating the ambiguity inherent in threshold-based diagnostics. The framework is validated not by forward clinical trials, but by "validation by impossibility": published statistics from ADNI (n=824), PPMI (n=423), UK Biobank (n=494,577), and Framingham (n=5,205) are fed through a branchless C++ engine. All heterogeneous inputs converge to the same attractor with maximum deviation 1.74×10⁻¹⁴. If the theorem holds, it is mathematically impossible for valid data to fail this test. Applications to Alzheimer's disease, Parkinson's disease, prion disorders, ALS, Huntington's disease, frontotemporal dementia, systemic amyloidoses, and type 2 diabetes. Includes complete source code (zero conditional branching), Phase 0/1 clinical validation protocol, and implications for AI alignment. Key theorem: π₁ = 0 ⟹ Unique Coherent Fixed Point.
