22 01 Base Architecture Clear Cell Renal Carcinoma ccRCC
Abstract
Clear cell renal cell carcinoma (ccRCC) accounts for 70-80% of renal cell carcinomas anddemonstrates remarkable metabolic flexibility contributing to therapeutic resistance. Thefounding molecular event—VHL tumor suppressor loss in 90% of cases—creates constitutiveHIF1α/HIF2α stabilization independent of oxygen availability . This pseudohypoxic statefundamentally reconfigures cellular architecture and metabolism.Current therapeutic approaches targeting VEGF pathways or immune checkpoints achievemodest response rates (30-40% ORR) with inevitable resistance emergence. This Model tries to paint a minimum picture of the base architecture and dynamics of this carcinoma.
