Ratio Imaging Analyzer (RIA): A Standalone, Zero-Dependency Tool for Robust Ratiometric Fluorescence Analysis
Abstract
Ratio Imaging Analyzer (RIA) is an open-source desktop application designed to streamline the processing of ratiometric fluorescence imaging data (e.g., FRET, Calcium, pH sensors). It bridges the gap between raw microscopy data and biological insight through a user-friendly Graphical User Interface (GUI). New in v1.8.3 - Dual-Track Architecture:To balance portability with professional power, RIA is now available in two editions: β‘ RIA Lite (Standalone EXE): A re-engineered, ultra-compact executable optimized for instant startup and maximum portability on standard Windows PCs, perfect for standard TIFF workflows without any installation. π RIA Pro (Python/PyPI): The full-featured Python version that integrates comprehensive dependencies to provide native support for professional microscopy formats, including Olympus .oir, Nikon .nd2, and Zeiss .czi. Key Features: π§ Robust Motion Correction: Integrates the Enhanced Correlation Coefficient (ECC) algorithm to automatically align image stacks and compensate for sample drift. π Scientific-Grade Signal Processing: Implements custom Normalized Convolution to handle NaN values correctly, preventing artifacts at image boundaries and masked regions during ratio calculation. β¨ High-Contrast ROI System: Features a custom multi-layered rendering engine for Regions of Interest, ensuring visibility against complex fluorescent backgrounds. π Analysis Unchained: Allows researchers to perform rigorous background subtraction, thresholding, and time-series extraction on personal laptops, freeing up restricted lab workstations.
