Abstract

sGP3 tetramer labelling of immunoglobulin G (IgG) B cells within peripheral blood mononuclear cells (PBMCs) from hyperimmune pig 64 (148 dpi) and a PRRSV-naïve pig (A). Back-gating of sorted single sGP3 tetramer-binding IgG B cells from pig 64 (B). Complementary DNA (cDNA) was synthesized from isolated single cells, and the variable region domains of IgG heavy (VH), Ig kappa (κ), and Ig lambda (λ) light chains (VL) were amplified; representative gel electrophoresis images are shown (C). Expected product sizes were 472 base pairs (bp) for VH chains, 477–512 bp for κ chains, and 422–477 bp for λ chains
