Abstract

Headspace (HS) sampling technique is widely used for the analysis of volatile and semi-volatile organic compounds. We analyzed the basic ideas, tools, and methodological variations of HS, such as the novel Solid Phase Dynamic Extraction (SPDE), Solid Phase Microextraction (SPME), and Static Headspace (SHS). The review summarizes the current uses of these methods in complex matrices, such as natural product profiling, pharmaceutical reaction monitoring, bisphenol analysis in water, and biomonitoring of n-heptane in blood. By comparing these "green" and solvent-less HS techniques with traditional and contemporary liquid-phase extraction methods, such as liquid-liquid extraction (LLE) for Persistent Organic Pollutants (POPs), Solid-Phase Extraction (SPE) for UV filters, Dispersive Liquid-Liquid Microextraction (DLLME) for forensic analysis, Matrix Solid-Phase Dispersion (MSPD) for biota, and direct solvent extraction for high-matrix oils, a central thesis is developed. We examine important developments, especially in-situ derivatization, which greatly increases the applicability of HS to polar and semi-volatile analytes. Lastly, based on a synthesis of the analytical performance metrics (Limits of Detection, Limits of Quantification, precision, accuracy, and matrix effects) reported across these groundbreaking studies, we develop a framework for method validation and reporting standards.
