Tracking Mitochondria in Murine Malignant Mesothelioma Cells using Confocal Raman Microscopy (Conference Abstract)
Abstract
Cancer cell plasticity drives therapy resistance and metastatic progression, supported by dynamic changes in organelle function, metabolism, and intracellular trafficking. Tools to non-invasively map these adaptive states are needed to understand how cancer cells reprogram organelle interactions and nanoparticle uptake. Confocal Raman microscopy (CRM) combined with bioorthogonal Raman tags (R-tags) offers a promising strategy for non-invasive, multiplexed imaging of cellular compartments. We aim to evaluate the suitability, performance, and cytotoxicity of an R-tag commonly used for mitochondrial labeling (MitoBADY), as part of a Raman-based framework to investigate the cellular microenvironment and facilitate the study of tumor cell - lipid nanoparticle (LNP) interactions.
