Hybrid mimetic Neu5Ac as a tool for tumor sugar shield collapse and NAD+ blockade.
Abstract
While current approaches focus on the enzymatic removal of the outer glycan layer, our strategy is based on integrative metabolic sabotage, turning the very process of building a sugar shield into a tumor death mechanism. Our new development is the use of 2-deoxy-D-glucose (2-DG) and 3-acetylpyridine in hybridization of GlcNAc - this is a serious step towards understanding how to "hack" the energy and protection of a cancer cell. This is true. In 2026, this approach – metabolic engineering of glycocalyx in combination with cofactor blockade – is considered the "aerobatics" of targeted therapy. We use the natural conveyor of a cancer cell so that it collects its "death" from our details. This turns the biochemistry of cancer into a tool for its own destruction. Our strategy using 3-acetylpyridine will induce local NAD+/NADH deficiency within the cancer cell, disrupting its metabolism – NAD blockade below the archaic tumor pathways:
