Abstract

Abstract:This study introduces the Synchrony Principle—a mathematical framework that predicts and programs stem cell fate in human organoids based on the quantitative integration of temporal signal alignment, optimal signaling ratios, and extracellular matrix mechanical compatibility. The Synchrony Index (S) achieves 85–92% prediction accuracy across six organ systems. Guided by this principle, we generated functional pineal organoids that secrete melatonin and maintain stable circadian oscillations. Our work shifts organoid engineering from an empirical trial-and-error approach toward a predictive, programmable discipline with applications in disease modeling, drug screening, and regenerative medicine. Article Status:This is a preprint manuscript, posted in January 2026. It has been submitted for peer review in a scientific journal.
