Abstract

Dataset Description: This dataset contains key experimental data for validating the mechanism that "MOV10 inhibits SADS-CoV replication by enhancing TRIM24-mediated K63-linked TRAF3 ubiquitination, and this inhibition is antagonized by the viral N protein." Detailed Methods Cells & Virus: The following cell lines and virus strain were used: Vero E6, HEK293T, ST, IPI-2I cells, and SADS-CoV. Key Experiments: The data encompasses results from the following procedures: Overexpression and knockdown of MOV10. Co-immunoprecipitation (Co-IP) assays for MOV10 with TRAF3 and the SADS-CoV N protein, as well as for TRIM24 with TRAF3. Western Blot analysis of MOV10 promoting K63-linked ubiquitination levels of TRAF3. Western Blot analysis of the SADS-CoV N protein promoting K48-linked ubiquitination levels of TRAF3. Data Collection: An Odyssey infrared imaging system (LiCor BioSciences, USA) was used to visualize the blots. File ContentsThis dataset comprises the following files: Wertern Blot Raw Data.docx: Original images of all Western Blots. PCR Raw Data.XLXS: Numerical data for all gene mRNA expression levels. Data Reuse & Attribution These data can be used directly to validate the conclusions of this study or for comparative analysis of coronavirus-host factor interactions. This dataset is directly associated with the preprint/manuscript (https://doi.org/10.1186/s13567-024-01303-z).
