Abstract

LFP Sleep Scoring & Ripple Analysis Pipeline This script comprises a pipeline to analyse LFP recordings from the dorsal hippocampus using an OpenEphys DAQ, with each tetrode targeting CA1, CA2, CA3, and DG. The implanted mice (10-12 weeks old) were allowed to explore their home cage for two hours (pre-social), followed by 5 minutes of Social interaction in the home cage with a novel, sex-matched juvenile mouse. After the social encounter, another 2 hours of home-cage exploration and recording were conducted. A trial dataset for running the script is provided (WT_tdTomato_pre.zip). Overview This Python script performs automated Local Field Potential (LFP) analysis for neuroscience data recorded via the Open Ephys acquisition system. It is designed to: Load Data: Parse Open Ephys files (structure.oebin, continuous.dat, timestamps.npy). Preprocessing: Downsample raw data (e.g., to 1 kHz) and extract specific channels based on region IDs. Sleep Scoring: Classify brain states (NREM, REM, Awake) using a PCA-based approach with thresholding (used in the paper) or a Delta/Theta power ratio. Ripple Detection: Detect Sharp-Wave Ripples (SWRs) in the CA1 region during identified NREM epochs using Z-scored power thresholds. Visualization: Generate average ripple spectrograms and Power Spectral Density (PSD) plots. Below is a technical description (as in Readme.pdf) of how to use the script.
