Abstract

This handbook compiles the laboratory protocols implemented for the isolation and characterization of mucins from sputum from asthmatic patients using proteomic analysis. It details the complete experimental workflow, including sputum samples solubilization, immunodetection of specific mucins via Western Blotting, and sample preparation for high-resolution mass spectrometry. Furthermore, the manual outlines the parameters for high-resolution mass spectrometry acquisition and the bioinformatic pipelines utilized for the processing and interpretation of mass spectrometry data. The document is openly shared to promote reproducibility and support future work in proteomic analysis of sputum samples for the identification of prognostic and diagnostic biomarkers of muco-obstructive diseases, like asthma.
