Abstract

The present study aimed to formulate and evaluate Telmisartan tablets using the solid dispersion technique to enhance solubility and dissolution rate. Telmisartan, a BCS Class II drug with poor aqueous solubility, was characterized for its physicochemical properties. The melting point of Telmisartan was found to be in the range of 261–264 °C, confirming its purity. UV spectroscopic analysis showed a λmax at 295 nm, and calibration curves exhibited excellent linearity with a regression coefficient (R² > 0.998). FTIR, DSC, and XRD studies confirmed the absence of drug–polymer interaction and indicated conversion of crystalline Telmisartan into an amorphous form in the optimized solid dispersion. Solid dispersions were prepared using Kollidon®12 PF and Eudragit® EPO by solvent evaporation method. SEM analysis revealed irregular porous morphology, indicating increased surface area. The optimized batch (A2) showed maximum solubility enhancement and drug content. Tablets prepared by direct compression demonstrated acceptable hardness, friability, weight variation, and disintegration time. In-vitro dissolution studies showed 94.46% drug release within 8 hours compared to 76.08% for plain Telmisartan, following zero-order kinetics. Stability studies for three months indicated no significant changes in physical properties or dissolution profile, confirming formulation stability. Overall, the solid dispersion approach proved to be an effective strategy to enhance the solubility, dissolution, and potential bioavailability of Telmisartan tablets.
